Winchell studierte an der Columbia University Medizin. Von 1950 bis 1954 war er Gastgeber der Fernsehsendung The Paul Winchell Show auf dem US-amerikanischen Fernsehsender NBC. Von 1965 bis 1968 war er Gastgeber der Kinderfernsehserie Winchell-Mahoney Time.
Winchell's first show as a ventriloquist was on radio with Jerry Mahoney in 1943. The program was short-lived, however, as he was overshadowed by Edgar Bergen . Winchell also created Ozwald , a character that resembled Humpty Dumpty .
In 1954, The Paul Winchell and Jerry Mahoney Show moved from primetime to Saturday mornings. Set in “Jerry’s Clubhouse,” the show now featured Winchell and Mahoney entertaining a live audience of children.
