After graduating from the normal school, Afanassjewa entered the Women's University in St Petersburg where she studied mathematics and physics under Orest D Chvolsou. In 1902 she went to Germany to continue her studies with Felix Klein and David Hilbert at the University of Göttingen. There she met Paul Ehrenfest .
Seit 1904 war er mit der russischen Mathematikerin Tatjana Ehrenfest-Afanassjewa verheiratet, die mit ihm in Göttingen studiert hatte und mit der er zwei Töchter (Tatjana van Aardenne-Ehrenfest, Galinka) und zwei Söhne (Paul Jr., Vassily) hatte. Ehrenfest, der an Depression litt, erschoss sich am 25. September 1933.
Tatjana van Aardenne-Ehrenfest wuchs in Sankt Petersburg auf, bevor ihre Eltern Paul Ehrenfest und Tatjana Ehrenfest-Afanassjewa mit ihr 1912 nach Leiden zogen. Paul Ehrenfest war ein bekannter theoretischer Physiker und Nachfolger auf dem Lehrstuhl von Hendrik Antoon Lorentz in Leiden, ihre Mutter ebenfalls bekannt als Mathematikerin und theoretische Physikerin.
